ABSTRACT

Background: Diabetes mellitus is a metabolic disease due to impaired insulin secretion and/or abnormalities in the action of insulin on target tissues. Many plants already demonstrated their efficacy in the treatment of this chronic disease. The present study was undertaken to evaluate the effects of the aqueous extract of Desmodium adscendens on insulin and glycemic reactivity in type 2 diabetic rats.

Methodology: Diabetes was induced by prolonged sucrose administration followed by a single injection of streptozotocin (STZ) at a dose of 40 mg/kg. Twenty-six diabetic animals were separated into four groups, receiving respectively distilled water (10 mL/kg), metformin (200 mg/kg) or aqueous extract of D. adscendens at 100 and 200 mg/kg by oral gavage during 28 consecutive days. A fifth group of 5 healthy rats received distilled water (10 mL/kg). During the experimentation, water consumption and body weight were evaluated. After 28 days of treatment oral glucose tolerance and insulin sensitivity tests were performed.

Results: Animals made diabetic by administration of sucrose and STZ showed a significant increase in the glycemia level (p < 0.001), water consumption (p < 0.001) and a non-significant reduction of weight loss compared to those of the healthy control group. Administration of the extract significantly (p < 0.001) decreased blood glucose and water intake compared to those of untreated diabetic rats. Moreover, administration of the extract at both doses significantly (p<0.001) enhanced insulin sensitivity, compared to untreated rats.

Conclusion: The results suggest that D. adscendens treatment was able to correct the increase in water intake and improve glucose metabolism.
